First, you cannot do a backup using fastboot. Fastboot is like a one-way road: you can only use it to write partitions (either by flashing a corresponding image, or by wiping it) – but you cannot "pull" a partition. Which leaves ADB. Yes, it's possible via ADB – but it will require root.

I have SmartLike Lyf Water 8 LS-5015 with Android 6.1 Marshmallow vendor updated. I want to take NAND backup before rooting through loading TWRP recovery in fastboot mode (fastboot recovery twrp.img). I tried with 3 TWRP recovery images listed below: one is TWRP 2.8.4 recovery image from the official TWRP website and the other two are from XDA.
If you modify your device, bottom line, you should leave it unlocked so you can fix things or go back to stock if needed, otherwise you could turn it into a paper weight. Even if root is done without modifying the boot.img, I would still keep it unlocked as there are usually still system changes.
